hiPSC-derived Dopaminergic Neurons

Human induced pluripotent stem cell derived dopaminergic neurons (DA) expressing target markers of substantia nigra pars compacta (SNc) residing dopaminergic neurons. These cells are knowns to be the most vulnerable population in Parkinson’s Disease (PD). A novel direct differentiation protocol was developed to differentiate human induced pluripotent stem cells to specialized A9 dopaminergic neurons. This protocol was developed through the application of HD-DoE at each stage of differentiation by screening more than 100 compounds in total. Therefore, generating a short-term protocol that targets upregulation of key transcription factors such as FOXA2 and LMX1A while minimizing stray fates in culture without relying on genetic reprogramming.
